Porcine intestinal lymphoid tissues synthesize estradiol

Abstract
17β-estradiol is synthesized primarily in the gonads of both sexes and regulates the development and function of reproductive organs. Lately, we discovered that intestinal lymphocyte homeostasis is regulated by estradiol synthesized de novo in the endothelial cells of high endothelial venule of mesenteric lymph node and Peyer’s patches in mice. This finding prompted us to hypothesize that the high endothelial venule of intestinal lymphoid tissues is the site of estradiol synthesis across the species. In this study, we determined if estradiol is synthesized in the porcine intestinal lymphoid tissues using adolescent piglets. Comparisons of the blood and tissue contents of estradiol showed that estradiol concentrations in mesenteric lymph node and Peyer’s patches were significantly higher than serum levels. RT-PCR showed that these intestinal lymphoid tissues express mRNAs for steroidogenic enzymes (StAR, 17β-Hsd, 3β-Hsd, Cyp17a1, and Cyp19a1) and immunohistochemistry in ilial tissue with a Peyer’s patch localized aromatase (CYP19) in the endothelial cells of high endothelial venule. When mesenteric lymph node and Peyer’s patches were cultured in vitro, they produced estradiol. Taken together, this study finds that mesenteric lymph node and Peyer’s patches are the sites of estradiol synthesis in the adolescent piglets.
